Biuret test: Biuret reagent is added to the liquid sample. In a positive test, copper II ion (Cu2+ ion) is reduced to copper I ion (Cu+ ion) which forms a complex with the nitrogen and carbon of peptide bonds in an alkaline solution. The presence of proteins is indicated by a violet colour.
